1H MRI study of small-diameter silk vascular grafts in water

چکیده
Small-diameter vascular grafts (VG) made from silk fibroin (SF) fibers by a double-raschel knitting method were prepared by coating their interior and exterior with different materials, such as SF or polyurethane (PU). The 1H spin density, 1H spin–spin relaxation time (T2) and diffusion coefficient (D) of the water molecules in the layer of the VG coated by these materials were non-destructively observed in water using 1H NMR imaging. The inner and outer coating materials significantly affected the amount and mobility of water molecules in the VG. The water content of the SF-coated VG was considerably higher than that of PU-coated VG. The T2 value of the water molecules in the SF-coated VG layer was smaller than that of the PU-coated VG, which means that the rotational motion of the water molecules in the layer of the SF-coated VG was restricted by the intermolecular interaction between the SF and water molecules. The diffusion coefficient (D) of the water molecules in the SF-coated VG layer was larger than that of the PU-coated VG, indicating that the water molecules were confined to the pores in the graft layer of the PU-coated VG. Histological Reactions and the In Vivo Patency Rates of Small Silk Vascular Grafts in a Canine Model
Objective: To evaluate in vivo patency rates of silk fibroin (SF) vascular grafts and resulting histological reactions in a canine model. Methods: To generate 3.5-mm inner diameter vessels, a combination of plaited silk fibers were wound with cocoon filaments and subsequently coated with an SF solution. The resulting SF grafts (n=35) were implanted into the carotid arteries of male beagles (age...
متن کاملSilk-based electrospun tubular scaffolds for tissue-engineered vascular grafts.
Electrospinning was used to fabricate non-woven nanofibrous tubular scaffolds from Bombyx mori silk fibroin using an all aqueous process. Cell studies and mechanical characterization tests were performed on the electrospun silk tubes to assess the viability of their usage in bioengineering small-diameter vascular grafts. Human endothelial cells and smooth muscle cells were successfully cultured...
متن کاملElectrospinning Bombyx mori silk with poly(ethylene oxide).
Electrospinning for the formation of nanoscale diameter fibers has been explored for high-performance filters and biomaterial scaffolds for vascular grafts or wound dressings. Fibers with nanoscale diameters provide benefits due to high surface area. In the present study we explore electrospinning for protein-based biomaterials to fabricate scaffolds and membranes from regenerated silkworm silk...
متن کاملDevelopment and in vivo validation of tissue-engineered, small-diameter vascular grafts from decellularized aortae of fetal pigs and canine vascular endothelial cells
BACKGROUND Tissue engineering has emerged as a promising alternative for small-diameter vascular grafts. The aim of this study was to determine the feasibility of using decellularized aortae of fetal pigs (DAFPs) to construct tissue-engineered, small-diameter vascular grafts and to test the performance and application of DAFPs as vascular tissue-engineered scaffolds in the canine arterial syste...
متن کاملPhysiological pulsatile flow culture conditions to generate functional endothelium on a sulfated silk fibroin nanofibrous scaffold.
Many studies have demonstrated that in vitro shear stress conditioning of endothelial cell-seeded small-diameter vascular grafts can improve cell retention and function. However, the laminar flow and pulsatile flow conditions which are commonly used in vascular tissue engineering and hemodynamic studies are quite different from the actual physiological pulsatile flow which is pulsatile in natur...
